目前醫院用來篩檢肝癌的方法為抽血檢驗,檢驗血清中的Alpha-fetoprotein(AFP),AFP的檢測靈敏度只有70%左右,但只憑AFP升高無法斷定罹患肝癌,還必須配合影像檢查,進行多種的檢驗來確認肝癌,多種檢驗造成效率低,固本研究研發披覆多重抗體試劑與混合試劑,以提高檢測之靈敏度。
我們將肝癌細胞種植於老鼠背部,肝癌細胞從老鼠體內獲取養分,然後形成腫瘤,記錄腫瘤成長過程,並且按照種植肝癌細胞後的0、1、3、5週,量測血清內的肝癌指標性蛋白,目的是為了觀察腫瘤在前期與後期的指標性蛋白表現量,我們使用Alpha-fetoprotein(AFP)試劑、Glypican-3(GPC3)試劑、Des-gamma-carboxy prothrombin(DCP)試劑、披覆多重抗體試劑及混合試劑來檢測,使用的方法為磁減量檢測法(ImmunoMagnetic Reduction, IMR),在後期我們將腫瘤取出,做成切片,以磁性奈米試劑進行磁染色,成功的用磁訊號觀察腫瘤指標性蛋白。
