隨著超高齡社會的來臨,高齡長者行動不便以及長期臥病在床等問題,已經造成醫療資源與健保費用的龐大支出,為緩解長者骨質疏鬆症和退化性關節炎,民間常以龜鹿二仙膠補強筋骨。本論文主要研究龜鹿二仙膠對於緩解關節與骨骼退化的替代醫療效果,本研究選用與順天堂科學中藥廠提供的龜鹿二仙膠,龜鹿二仙膠主要藥材包括枸杞、龜板、人參和鹿角。本研究首先利用1,1-二苯基-2-吡啶並肼基(1,1-diphenyl-2-picrylhydrazyl;DPPH)自由基清除能力測試檢視龜鹿二仙膠的抗氧化力;再利用脂多醣(lipopolysaccharide,LPS)處理關節滑膜細胞(HIG-82),藉此誘發關節滑膜細胞(synoviocyte,HIG-82)的發炎現象,透過細胞存活率分析法(3-(4,5-Dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide;MTT assay)以及酵素結合免疫吸附分析法(Enzyme-linked immunosorbent;ELISA assay),檢視龜鹿二仙膠是否可以緩解脂多醣(LPS)誘發關節滑膜細胞(HIG-82)的發炎;此外,本研究利用化療藥物阿黴素預先處理,藉此誘發老化小鼠的骨骼疏鬆,然後使用骨骼電腦斷層掃描技術檢視脛骨的骨小樑與骨密度,藉此評估龜鹿二仙膠是否可以緩解化療藥物阿黴素所誘發小鼠的骨質疏鬆。本論文結果顯示龜鹿二仙膠不但可以顯著增強DPPH自由基清除能力,對於脂多醣(LPS)誘發關節滑膜細胞(HIG-82)的發炎,也有顯著的緩解效果;此外,對於化療藥物阿黴素(doxorubicin,DOX)所誘發小鼠的骨質疏鬆,連續餵食龜鹿二仙膠四週後,也有顯著的緩解效果。歸納本研究結果,可以說明龜鹿二仙膠對於緩解關節與骨骼退化確實具有替代醫療效果。研究者認為適時給予中、高齡長者補充龜鹿二仙膠,將有助於緩解高齡長者行動不便以及長期臥病在床等問題,大大降低醫療資源與健保經費的龐大支出。
With the advent of the super-aged society, elders' problems such as mobility problems and long-term bedridden cause huge expenditures on medical resources and health insurance costs. In order to alleviate the osteoporosis and degenerative arthritis of the elders, the people often use the Guilu Erxian Glue (GEG) to relieve the osteoporosis and degenerative arthritis. This thesis mainly studied alternative medical potential of the GEG in relieving joint and bone degeneration. The GEG, provided by Sun Ten Pharmaceutical Co., Ltd., with four main ingredients of ginseng, wolfberry, tortoise plastron and antler from a famous Chinese prescription. In this study. (we used 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay to examine the antioxidant capacity of the GEG; then we used lipopolysaccharide (LPS) to treats HIG-82 joint synovial cells, thereby inducing the inflammation in HIG-82 cells, through the 3-(4,5-Dimethylthiazol- 2-yl )-2,5-diphenyltetrazolium bromide (MTT) and enzyme-linked immunosorbent assay (ELISA) to examine whether the GEG can alleviate chemotherapy-induced HIG-82 joint synovial cell toxicity and Inflammation. In addition, we used the doxorubicin (DOX) to induce bone loss in aging mice, and then used bone computer tomography to examine the trabecular density of the tibia, thereby assessing whether the GEG can alleviate the osteoporosis in chemotherapy mice. Our results of this thesis showed that GEG could not only significantly enhance the DPPH free radical scavenging ability, but also had a significant alleviating effect on the toxicity and inflammation of chemotherapy HIG-82 cells. In addition, After feeding GEG for four consecutive weeks, those chemotherapy mice had a significant relief effect in osteoporosis. To summarize our results, the GEG indeed have an alternative medical effect on alleviating joint and bone degeneration. We believe that timely supplementation of GEG to the middle-aged people that should help to alleviate the mobility problems and long-term bedridden problems for elders, and greatly reduce the huge expenditures on medical resources and health insurance costs.
