高山芒(Miscanthus transmorrisonensis)是台灣的特有物種,分布在台灣中高海拔2200公尺以上的山區,為高山草原的主要組成之一,一般認為它是靠風力傳播的物種,但該物種的遺傳變異及族群分化至今尚未明瞭。本實驗係利用同功酵素方法檢測高山芒的族群變異度及其遺傳結構。族群樣本共得自合歡山、雪山、大雪山、南湖大山、玉山、關山、關山嶺、塔關山等八個族群。總共分析了九種同功酵素,判讀了16個基因座,其中具有多型性的基因座有12個。各族群的平均多型性基因座百分比為56.3%,各基因座上的平均對偶基因數接近2個,各族群的異合度(HE)平均值為0.21,而異合度觀察值(HO)平均為0.17,這些數值顯示高山芒的族群遺傳變異度豐富。此外,計算遺傳變異的分布情形,可發現高山芒的遺傳變異度僅有10%是存在於族群間(Gst=0.10),90%存在於族群內,基因流傳尚屬通暢,另外,高山芒各族群之間遺傳相似度平均值為0.97,這表示高山芒的族群間略有遺傳分化。整體而言高山芒所表現的遺傳結構符合一般風媒物種的特性。
Miscanthus transmorrisonensis Hayata is an endemic gramineous species of Taiwan .Genetic variation within and among populations were determined by isozyme electrophoresis analysis. Samples were collected from 8 different populations: Ho-huan shan, Hsueh shan, Ta-hsueh shan, Nan-hu ta shan, Yu shan, Kuan shan, Kuan-shan Ling and Ta-kuan shan. Allozyme frequency analysis of 16 loci showed the mean percentage of polymorphic loci is 56.3%, mean alleles per locus approximates 2, mean expected value of heterozygosity ( HE ) is 0.21 and mean observed value of heterozygosity ( Ho) is 0.17. All those preceding figures suggest that population genetic variation exists in M. transmorrisonensis. Meanwhile, it was found only 10% of genetic variation existed among populations, and the rest 90% existed within populations.Moreover, the genetic similarity among M. transmorrisonensis populations is 0.97 . This result is in accordance with general characters of wind-pollinated species.
